A method has been designed for electrophoretic transfer of proteins from polyacrylamide gels to nitrocellulose sheets. The method produces quantitative transfer of ribosomal protein gel containing urea. For sodium dodecyl sulfate gels, the original band pattern is obtained without loss of resolution, but the transfer is not quantitative. This method allows the detection of proteins by autoradiography and simpler than conventional procedures.
The immobilized proteins were detected by immunological procedures. All additional binding capacity on the nitrocellulose was blocked with excess protein; then the specific antibody bound and, finally, a second antibody directed against the first antibody.
The second antibody either radioactively labeled or fluorescein or peroxidase conjugate. Certain proteins are then detected by either autoradiography, under UV light, or with a peroxidase reaction product, respectively.
In the latter case, as little as 100 pg of protein clearly detectable. It is anticipated that the procedure will be applicable to the analysis of various proteins with a particular reaction or a ligand.
Dystrophin: the protein product of the Duchenne muscular dystrophy locus.
The protein product of the human locus Duchenne muscular dystrophy (DMD) and its mouse homologue (mDMD) have been identified using polyclonal antibodies directed against fusion proteins that contain two different areas of mDMD cDNA. DMD protein proved to be about 400 kd and represent approximately 0.002% of the total skeletal muscle protein. These proteins were also detected in smooth muscle (stomach). muscle tissue isolated from both boys affected by DMD and mdx mice DMD no protein is detected, indicating that this genetic abnormality homologous.
Since mdx mice present a clinical disorder is not clear, the identification of mdx mouse as an animal model for DMD have important implications with regard to etiology DMD lethal phenotype. We have named dystrophin protein for identification through isolation Duchenne muscular dystrophy locus.
Generation of a large number of dendritic cells from mouse bone marrow cultures are equipped with granulocyte / macrophage colony-stimulating factor.
Antigens, major histocompatibility complex (MHC) class II-rich dendritic cells known to arise from the bone marrow. However, marrow less mature dendritic cells, and a large number of proliferating immature cells have not been identified. Methodology to encourage the growth of dendritic cell that was recently described for the mouse blood has now been modified to MHC class II-negative precursors in the marrow.
A key step is to remove the majority of nonadherent, newly-formed granulocytes by gently washes during the first 2-4 d culture. It leaves behind the breed cluster loosely attached more firmly adherent “stroma.” At 4-6 days the cluster can be dislodged, isolated by 1 g sedimentation, and after reculture, large numbers of dendritic cells are released. The latter can be identified on the basis of different forms of cells, ultrastructure, and a repertoire of antigens as detected by the monoclonal antibody panel. Dendritic cells express high levels of MHC class II products and act as a powerful accessory cells to initiate leukocyte reaction mixture.
