host cell protein (HCP) which is related to the process of manure produced by the host organism and is typically present at low levels in recombinant biopharmaceutical products, such as therapeutic antibodies. While the overall level of HCP is usually monitored by enzyme-linked immunosorbent assay (ELISA), liquid chromatography coupled with mass spectrometry (LC-MS) is emerging as a powerful tool that can provide both qualitative and quantitative information about the level of HCP during the purification process development. However, a big challenge for LC-MS-based method is that there can be more than 5 orders of magnitude difference in concentrations between HCP and therapeutic antibodies in solution, which hinders effective identification of low abundance HCP in the product antibodies.
This work reports a simple and powerful strategy to identify HCP in antibody drug substance by implementing measures molecular weight cut-off (MWCO) filtration followed by shotgun proteomics analysis. After separating the interaction between HCPs and antibodies with anionic detergent, antibody depletion of HCP can be easily achieved with MWCO filtration step. By using this method, we observed that the dynamic range in the protein in the sample HCP significantly decreased to 1,000-fold. In addition, by spiking known amounts of HCP to purified antibody drug substance with low levels of HCP, we show that our method can detect HCP are low molecular weight (11 kDa and 17 kDa) at concentrations as low as 1 ppm.
When applying this methodology to study at NIST HCP monoclonal antibodies (NISTmAb), more than 150 HCPs who confidently identified, which doubles the amount of HCP identified previously reported. Parallel reaction monitoring (PRM) results confirms that the new HCP is found using this method are present in very low abundance (0.01 to 8 ppm), highlighting that our method reduces the dynamic range by removing the antibody interference and increases sensitivity HCP identification and quantification.

Here, we describe a new technology to modulate translational strength chain antibody through a Kozak sequence variants to generate bispecific antibodies in a single cell line. In this study, we designed and screened a large Kozak sequence library to identify 10 variants that can independently modulate protein expression levels of about 0.2 to 1.3-fold compared with the sequence of Wild-type transient transfection.
We use a combination of several variants, covering a range of translational strengths, to develop a single cell CHO cell lines bispecific stable and compared the results with those obtained from wild sequence types. A significant increase in the assembly of bispecific antibody with a concomitant reduction in the level of product-related impurities were observed.
